ABSTRACT

OBJECTIVETo explore the effects of Osthole on apoptosis of HL-60 cells induced by tumor necrosis factor related apoptosis inducing ligand (TRAIL) and its possible mechanism.

METHODSThe proliferative inhibition of HL-60 cells treated with different concentrations of Osthole, TRAIL alone and Osthole combined with TRAIL was measured by MTT assay. The HL-60 cells were treated with Osthole, TRAIL alone and Osthole combined with TRAIL at the concentration<ICvalue, i.e. 100µmol/L for Osthole and 40 ng/ml for TRAIL. Apoptosis and mitochondrial membrane potential (MMP) of HL-60 cells were detected by flow cytometry; the mRNA expression of BCL-2, BAX and DR5 was determined by RT-PCR; and the levels of Caspase-3,-8,-9 activity were detected by spectrophotometry.

RESULTSThe combined treatment (100µmol/L Osthole + 40 ng/ml TRAIL) of HL-60 cells for 48 h induced an apoptotic rate of (33.9±2.7) %, which was significantly higher than that of cells treated with Osthole or TRAIL alone (P<0.05); at the same time, the combined treatment promoted the decrease of MMP and the expression rate of BCL-2/BAX, and potentiated the expression of DR5 and Caspase-3,-8,-9 activity.

CONCLUSIONOsthole can sensitize HL-60 cells to TRAIL-induced apoptosis, which may be related with the activation of mitochondrial pathways and up-regulation of DR5.

ABSTRACT

OBJECTIVETo provide the basic guidance for seed breeding and cross-breeding of Paris polyphylla var. yunnanensis.

METHODThe floral behavior and pollinators were observed; 0.5% TTC solution was used for the pollen viability test and benzidine and -H2O2 was used for estimation of the stigma receptivity. The mating systems were tested by out crossing index (OC1), and pollination experiment was carried out by bagged and emasculated test in the field.

RESULTCommonly, stigma lobes spread slightly, and anthers started presenting the pollen from the outer ring while the flower was just beginning to open. Consequently, the distance between the stigma and its own pollen was relatively far, this "floral behavior" may be conducive to outcrossing. Then the flower entered the later period, while the stigma lobes spread widely, anthers all split, and this "floral behavior" shortened the stigma and its own pollen's distance, which may be conducive to selfing. P. polyphylla was partly protogynous. Stigma life-span was about 10-12 d. After anther dehiscence, the pollen viability maintained about 10% within 2 days, and 20% within 10 days. The value of out crossing index (OC1) was 4. By pollination experiment and pollinators observed, P. polyphylla was self-compatible, but no capacity for autonomous self-fertilization; In natural circumstances, outcrossing fructification rate was low, and mainly anemophilous. Assisted selfing-fertilization fructification rate was higher, spider was the main pollinators.

CONCLUSIONP. polyphylla has a mixed mating system with self-pollination and cross-pollination characteristics. Floral behavior has important adaptive significance in avoiding female and male interference, outcrossing, and delayed selfing. P. polyphylla is ambophily (a combination of both wind and insect pollination), pollinators changes due to environment. Pollen limitation is the main cause of low fructification rate under natural conditions.

ABSTRACT

OBJECTIVETo observe therapeutic effect of Shaoshanhuo needling method on periarthritis of shoulder.

METHODSOne hundred and twenty cases were randomly divided into two groups: a treatment group of 57 cases and a control group of 63 cases. The treatment group were treated with Shaoshanhuo needling method and massage combined with functional exercise of shoulder joint, and the control group were treated with warming acupuncture and moxibustion combined with massage and functional exercise of shoulder joint. Their therapeutic effects were evaluated after 2 courses.

RESULTSThe effective rate and the cured rate were 96.5% and 70.2% in the treatment group, and 88.9% and 50.8% in the control group respectively, with no significant difference between the two groups in the effective rate (P>0.05), but with a significant difference between the two groups in the cured rate (P<0.05), the treatment group being higher than the control group.

CONCLUSIONShaoshanhuo needling method has a better therapeutic effect on periarthritis of shoulder.

ABSTRACT

Objective To construct an expression system to produce the virus-like particles containing a part of the sequence of PSA mRNA, which are ribonuclease-resistant due to the encapsulation of the mRNA by bacteriophage MS2 coat proteins. Methods The PCR products of PSA cDNA fragments were cloned to TA vector pBS-T, then the targeted segments could be obtained when the pBS-T-PSA were digested by restriction endonuclease Hind Ⅲ and cloned to prokaryocytic expression vector pNCCL1. The recombinant plasmids named PNCCL1-PSA were transfected into E. Coli BL21-DE3 and induced to express with IPTG. Results The recombinant plasmids were successfully constructed. The bacteriophage MS2 coat protein which expressed in BL21 can self- assemble to form ribonuclease resistant virus-like particles and the PSA mRNA was encapsulated into virus-like particles. Conclusions The virus-like particle containing PSA mRNA can be expressed in prokaryocyte and it can be used as standard and control in detecting PSA mRNA. It provides a new, stable and ribonuclease-resistant RNA standard in RNA detection.
